U1148- TRANSLATIONAL VASCULAR RESEARCH LABORATORY (LRVT)

MISSIONS

  • We are recruiting an engineer to join a breakthrough project on cerebral aneurysms. Approximately 15 million people in Europe live with unruptured cerebral aneurysms, which can rupture suddenly, causing subarachnoid hemorrhages. Rupture often leads to death or severe disabilities, and current treatments are invasive, especially when aneurysms are asymptomatic. This leaves clinicians facing a difficult choice: operate or monitor. Unfortunately, safe non-invasive treatments do not exist, as we do not yet understand why some aneurysms rupture and others remain stable. Research from our laboratory shows that platelets, blood cells involved in thrombosis, play a key role in the development and rupture of aneurysms. Our initial findings indicate that platelets activate and accumulate at aneurysm sites before rupture, with a molecule, PF4, which could contribute to this process. To deepen these findings, we have formed a multidisciplinary and international team (France, Germany, and the Netherlands) to understand how platelets and PF4 influence aneurysm progression. Through advanced imaging, preclinical models, and genetic analyses and human studies, our goal is to determine if PF4 administration could offer a non-invasive treatment to prevent aneurysm rupture.

MAIN ACTIVITIES

  • Plan and perform experiments to test PF4 administration in a murine model of intracranial aneurysm. The execution of the project involves: - stereotaxic surgery, Ultrasound Doppler imaging and MRI, management of mouse colonies - planning and performing experimental models in mice - drafting/amending APAFIS projects - tissue analysis by different methods: biochemical assays (ELISA), flow cytometry, immunohistochemical staining on thin sections and whole tissues, participation in the formatting and presentation of generated results.

ASSOCIATED ACTIVITIES

  • Process and analyze data, format results for their presentations
  • Write experiment or study reports, protocols, and technical notes.
  • Keep the electronic laboratory notebook (Labguru) up to date
