The engineering assistant will join the 'Mécanismes physiologiques te pathologiques du développement cortical' team led by Juliette Godin.
We are looking for a motivated and curious engineering assistant in biological experimentation to join the "Mécanismes physiologiques et pathologiques du développement cortical" team at the Institut de Génétique et de Biologie Moléculaire et Cellulaire (IGBMC) in Strasbourg, France.
Within an international, interdisciplinary, and dynamic scientific environment, our team seeks to understand the fundamental mechanisms that control the acquisition of cell fate and the maturation of neurons during corticogenesis in mammals. The candidate will join a team working at the interface of neurobiology, molecular biology, and genomics, and will contribute to an ambitious research project funded by the European Research Council (ERC).
The project aims to understand how the modulation of the abundance and/or modifications of transfer RNAs (tRNA) influences translation and, more broadly, neuronal differentiation programs. The candidate will play a key role in the experimental implementation of this project.
The main mission will consist of generating and analyzing sequencing libraries to characterize the abundance and modifications of tRNAs, as well as developing and performing ribosome profiling experiments.
The candidate will also contribute to the team's experimental activities through various approaches, including:
- histology and immunohistochemistry on brain sections;
- biochemistry, notably Western blot;
- molecular biology, including PCR and qPCR;
- management and monitoring of several mouse colonies, in connection with the team's projects.
The position thus offers the opportunity to develop or strengthen diverse technical expertise, while participating in an exciting scientific project.
At the interface of several projects and in close interaction with different team members, the candidate must demonstrate rigor, autonomy, and excellent organizational and communication skills. Particular attention will be paid to the quality of experimental monitoring, laboratory notebook maintenance, reporting of results, and sharing information within the team.
MAIN ACTIVITIES:
- Development of tRNA libraries (mimtRNAseq) and ribosome profiling
- Cell culture (immortalized lines, murine primary cultures): maintenance of lines, transfections, optimization of a differentiation protocol from neural precursors into neurons)
- Management of mouse lines (Genotyping and colony management)
- Dissections of embryonic and adult murine brains
- Histological sections using cryostat, vibratome, microtome
- Immunostaining, confocal microscopy, cell counting (Qupath, ImageJ)
- Protein extractions, western blots, RT-qPCR, PCR
- RNA extractions by various methods
- Transmission of know-how and techniques
- Management of common areas and consumable stocks
- Oral and written experimental activity reports
